You cannot OD on one pill. There's no drug that you could find in a pill that will make you OD on 300mg (which is about the average weight of a pill).
You cannot OD on MDMA in any easy to take/obtainable capacity. You'd have to take quite a few grams of pure MDMA for it to kill you.
Be careful, though, pills today are rarely ever clean, and the other stuff in them can be dangerous. Popping a few pills of meth or speed bombs won't kill you, but it can make you pay like hell the next day. There's also a drug called PMA which produces rolling-like effects at low doses, but also has a much lower active dose (30mg for effects, 60mg to OD; this is the only drug that can kill you with one pill, and it's extremely rarely found now).
If you know what's in your pill, and you're willing to take the risks associated with that chemical, drop 2 and you will be good for the whole night. If you don't know what's in your pills, don't drop. There's a lot of bad stuff going around right now that will give you a horrible night (not kill you, but it's still bad for you).

Its possible if its a dirty pill, its rare that people od from one pill most deaths are caused by dehydration or heart problems. I would stay away from E if I was you. Causes allot of problems with depression, anxiety, concentration problems and memory.
